ABSTRACT: Physiological function requires modulated communication across the cell membrane, mediated in part by proteins that span the membrane with transmembrane segments. How the function, organization and drug interactions of these proteins involve their membrane environment, has been quantitatively studied with increasingly refined mathematical models over the last three decades. In my talk, I will present the progressive development of these mathematical models. I will focus especially on recent methodological developments from my work. These have identified and incorporated essential molecular details of structurally complex transmembrane proteins into the quantitative evaluation of their interactions with the membrane. This was made possible by the development of a novel hybrid method that combines a molecular description of the transmembrane proteins with an elastic continuum description of the membrane. In this hybrid method, the boundary conditions in the boundary value problem for membrane deformations incorporate information from molecular dynamics simulations of the transmembrane protein. Illustrative applications shed new light on the involvement of the membrane in the function and organization of primary drug targets like G-protein coupled receptors and transporters, including in their interaction with different drugs.
